Phage display nanobody selection from our fully synthetic humanized library [1] is a reality with more than a hundred successful projects since the publication of our library [2-7].
To complete our phage display selection of nanobodies, we took advantage from our 20 years of experience in Yeast genetics and developed both a Yeast 2-Hybrid platform and a Yeast display platform for optimization and intrabodization of nanobodies coming from our library or from the lama [7].
The selection and optimization of nanobodies and specifically nanobodies that could be expressed inside the cells (Intrabodies) will be illustrated with Nanobodies directed against TAU and providing anti-aggregative activity in vitro, in cellulo and in vivo in a mouse model of tauopathy.
